The bronchodilator tiotropium bromide, also known as tiotropium bromide, is a new type of bronchodilator successfully developed by the Dutch company Boehringer Ingelheim. It is a long-acting anticholinergic drug that has selective inhibitory effects on M3 receptors. , has the advantages of long blood concentration maintenance and minimal side effects. Tiotropium bromide is as effective as ipratropium bromide in the treatment of chronic obstructive pulmonary disease (COPD), but its advantage is that it only needs to be administered once a day. , while the latter requires administration 4 times a day. Recent studies have shown that long-acting β2-adrenoceptor agonists (such as formoterol) can dilate the bronchi equally effectively as short-acting ones. Both formoterol and tiotropium bromide can significantly increase forced expiratory volume in one second (FEV1), forced vital capacity (FVC) and resting deep inspiratory capacity (IC), and the combination of the two is more effective than either alone. Lung function improved more significantly.

production method:

50.87g methyl bis(2-thienyl)glycolate (I) and 31.04g hyoscyamine (II) were dissolved in 100ml toluene, 1.65g metallic sodium was added in batches at 90°C, and then heated at 78-90°C and 50kPa , distilled for 5h to remove the generated methanol. After treatment, 33.79g of compound (III) was obtained. Then in a mixture of dichloromethane and acetonitrile, react with methyl bromide for quaternization to obtain the product.
